Preparing X-Ray Crystallography Samples

Users should provide liquid protein or protein complex samples with the protein concentration of ~10 mg/ml or higher and purity of 95% or higher (gel and protein spectrum should be included). To remove possible aggregation products the sample should be cleared by centrifugation (30 minutes run at the speed of 13000 r.p.m.).

For successful crystallization it is essential that the protein or protein complex has a homogeneous oligomerization state. To maximize the chances of success for your project it is recommended to check the aggregation state of the protein or protein complex beforehand using one of the following techniques:

  • Gel-filtration
  • Dynamic Light Scattering
  • Analytical Ultracentrifugation
